A Dual Ionic pH-senstive copolymer for protein delivery |

The polymeric complex has been proved as an effective strategy for gene and protein delivery. The complexes from cationic polymer and anionic DNA/protein can maintain a stable state through electrostatic interactions in an aqueous medium. A dual ionic pH-sensitive copolymer was made for developing efficiency of protein delivery using such advantages. A copolymer poly(urethane amino sulfamethazine)(PUASM) was prepared by the urethane reaction. PUASM shows dual ionic pH-sensitive property in aqueous solution. At pH 8.5, it shows negative charge and soluble in water, and Lysozyme is also soluble but positive charged. When decrease pH to 7.4, PUASM can form micelle with loading lysozyme. At pH 5.5, there is demicellization of polymeric micelle. The polymer molecules changed to positive charge and release the Lysozyme. This dual ionic pH-sensitive copolymer can be used as effective protein carrier for ischemia stroke or many other diseases. |
