Engineering Soluble PD-1 for Improved Anti-Cancer Therapy |

Although therapeutic antibodies, which inhibit the interaction between PD-1 (Programmed cell death protein 1) and PD-L1 (Programmed cell death-ligand 1), have revolutionized in the field of immuno-oncology, they have a significant limitation with poor tumor penetration. To address the issue, we engineered the ectodomain of PD-1 using bacterial display and flow cytometric high-throughput library screening, and a variety of novel PD-1 variants exhibiting improved binding affinity to PD-L1 were successfully isolated. After production of the resulting novel PD-1 variants in mammalian cells, their various physicochemical and biological functions, including T cell activation, were thoroughly characterized. The isolated PD-1 variants will provide a promising platform for various pharmaceutics for PD-1 / PD-L1 blockade. |
