제목 Bone formation on polymer/bioceramic composites made by Baking-and-Press method
초록 There are many studies about preparing polymer/ceramic composites because of their good formability. However, most processes using solvent have the problem of decreasing the effect of bioceramic as an osteogenic induction factor due to the hindering of bioceramic by polymer. The purpose of this study is to fabricate polymer/ceramic composites by a press-and-baking method for rapid bone regeneration through enhancing the interaction of cells and bone induction factor. The scaffold was prepared with polylactic acid(PLA) as a main matrix, calcium metaphosphate as a inductive growth and differentiation factor and NaCl as a porogen for a 3-dimensional structure. Thereafter, osteoblasts from rat calvarias were seeded and cultured in vitro for 5 weeks. Also, the osteoblasts-seeded scaffolds were implanted at the backs of nude mice subcutaneously to investigate their biocompatibility. Cell-scaffold constructs were evaluated with MTT assay, DNA assay, and alkaline phosphatase activity measurement. In addition, SEM and histological study were performed to observe a tissue development. Cells were better grown to form bone-like structures on scaffolds fabricated by the press-and-baking method than cells in scaffolds fabricated by solution casting method. Also, the result of alkaline phosphatase activity assay showed that osteogenic differentiation was sustained and enhanced significantly in the scaffolds fabricated by the press-and-baking method.
