Stiffness analysis of synthesized protease degradable hydrogel based on hyaluronic acid |

In this presentation, we proposed a method for preparing hydrogels degradable by alpha-chymotrypsin. Protease initiated degradation was revealed by the stiffness analysis before and after treating protease. The protease degradable hydrogel is composed of acrylated hyaluronic acid crosslinked by a peptide crosslinker containing a proteolytic degradation site. Proteolytic degradation site is a peptide that sequence ‘CYKC’ is cleavable by alpha-chymotrypsin treatment. Two thiol groups from terminal cysteines can be crosslinked with acrylated hyaluronic acid by thiol-ene click reaction. The protease degradation of hydrogel was monitored by measuring stiffness changes. We envision that these protease degradable hydrogels are useful for tissue engineering and drug delivery. |
