In this study, injectable, thermo-sensitive, tissue adhesive hydrogels inspired by natural materials were designed. Amine-catechol rich chitosan combined with thiol-terminated pluronic F127 have potential to adhere to tissue. Hydrocaffeic acid conjugated chitosan (Chi-C) and thiolated pluronic F127 (Plu-SH) were mixed to make tissue adhesive hydrogels via bio-inspired catechol-thiol interactions and catechol-amine interactions. Chi-C and Plu-SH were synthesized and characterized using 1H NMR and UV-vis spectroscopy. Physical and mechanical properties of mixed Chi-C/Plu-SH hydrogels were evaluated using rheometer. We also confirm in vitro and in vivo stability by mass erosion rate of these hydrogels under physiological conditions. Using rheometer, universal testing machine (UTM), and hemostatic rat model, the tissue adhesion properties of the Chi-C/Plu-SH hydrogels were evaluated. |
