Light-response associated drug delivery has been studied a lot for cancer treatment. However, the light-response drug delivery declines efficacy for cancer therapeutics and unexpectable side effects. In this study, we designed photo-stimulated drug vehicle (PSDV) respond by tumor hypoxia and light stimuli. An anti-cancer drug loaded PSDV was assembled itself by reactive oxygen species (ROS)-mediated truncated polysaccharides conjugated with a photosensitizer. In hypoxia, ROS was generated by the electron rich in the polysaccharide chains. The ROS generated by PSDV allowed drug release at low oxygen condition in laser irradiation. Drug loaded PSDV showed toxicity in cancer cell line under hypoxia than that normoxia. In vivo and ex vivo experiments indicated that PSDV accumulated in the tumor region specifically. This research could be effective methods for treatment of cancer and hypoxic conditional diseases. |
