Biodegradable polymers have been used to develop cardiovascular prostheses such as vascular grafts and stents. However, thrombosis, inflammation, and restenosis that are associated with implant are still major obstacles for the use of these devices. In addition, several clinical reports demonstrated that some inflammatory markers exhibited relative rise in the first 10 minutes after inserting stent. We examined the expression levels of IL-6 after treating smooth muscle cell (SMC) and endothelial cell (EC) with the degradation product of various polymers and sirolimus (SRL) to evaluate the inflammation caused by acidic byproduct of various biodegradable polymers. Lactic acid, one of product of degraded polymers, induced inflammation and cell death. SRL played a role as anti-inflammatory reagent in limited range. The results demonstrated the feasibility of approaches to deliver anti-inflammatory therapeutic agents. |
