Decellularized Nanofibrous Scaffolds for Bone Tissue Engineering |

Scaffolds serve a key role in improving tissue regeneration by stem cells. Here, we report a decellularized tendon-derived scaffold with nanofibrous structures, which can enhance osteogenic differentiation and bone formation of human adipose-derived stem cells (hADSCs). Decellularized tendon scaffolds induce highly enhanced alignment and extension of hADSCs, leading to enhanced osteogenic differentiation of hADSCs in vitro. Transplantation of hADSCs using tendon scaffolds significantly promoted bone formation in a critical-sized calvarial bone defect model. Stacking the scaffolds seeded with hADSCs and human endothelial progenitor cells (hEPCs) further enhanced in vivo bone regeneration. In this study, we suggest a biomimetic tendon scaffold providing an efficient biomaterial platform for bone tissue engineering.
