Scaffold-free delivery of multi-layered cell assembly via thermally expandable hydrogels presenting controlled cell-adhesive property |

Scaffold-free engineering of 3 dimensional (3D) multi-layered cell assembly has been pursued in regenerative medicine. Despite various attempts, there are still some limitations such as long process time and laborious steps. In this study, we developed thermally expandable hydrogels which can deliver multi-layered cell assembly in a rapid and simple manner. We induced 3D cellular interactions by coating human dermal fibroblasts with fibronectin (FN) and gelatin in layer-by-layer assembly. We then coated the hydrogel with polydopamine and sequentially with FN to control cell-substrate binding. Multi-layered cell assembly was formed on the developed surface and readily detached with high efficiency (>90%) within 10 min mediated by expansion of the hydrogel at 4 oC. The assembly was transplanted to subcutaneous region of mouse, which showed retention for 14 days with improved angiogenesis. The platform can be applied to applications such as drug screening and regenerative medicine. |
