제목 Preparation and Characterization of Thermo-Responsive Polymer Surface asNon-Invasive Stem Cells Culture/Recovery Substrates
초록 We usually have been used trypsin-EDTA as breakdown enzyme of proteins during cell culture for subculture in vitro. However trypsin-EDTA can damage to not only adhesive proteins between cells and matrices but also cell membrane proteins. For this reason, we hypothesized that the damaged cells by trypsin-EDTA are dissatisfied with adhesion, proliferation, migration and differentiation in vitro and in vivo. In this study, we prepared thermo-responsive poly(3-hydroxybutyrate-co-3- hydroxyvalerate) (PHBV) nanofiber surfaces to recover intact cultured cells. The PIPAAm was grafted by electron beam irradiation on the surface of PHBV nanofiber mats to achieve non-invasive stem cell culture/recovery. When the temperature is reduced below 32℃, the PIPAAm-grafted PHBV surface becomes hydrophilic, allowing ADSCs to detach from the culture surface without enzyme treatments such as trypsin-EDTA.
