Anticancer potential of Metasequoia glyptostrobodies cones extract against human cervical cancer cells in vitro |
초록 |
This study is development to anticancer effect of Metasequoia glyptostroboides cones. Metasequoia glyptostroboides is known for its antioxidant and antifungal effects. Metasequoia glyptostroboides was extracted by methanol and partitioned by liquid-liquid extraction (Hexane, Dichloromethane, Chloroform, Ethyl acetate, and Water). HeLa cell was used as a cancer model and COS-7 cell was used as a normal model. Anticancer effect was evaluated by using cell counting kit-8 (CCK-8) assay. To characterize partitioned samples, analysis of total phenolic content, DPPH Assay, and DCF-DA assay were performed. Cell cycle analysis using Cellometer was performed to confirm the differences between components of the cell cycle. And then, qRT-PCR and western blot were used to specifically analyze the cell death pathway by measuring the relative quantities of cell death pathway-related genes and proteins respectively. |
