Release of Paclitaxel (PTx) loaded biodegradable MPEG-PLGC films for drug-eluting stents |

To use as drug eluting stent, preparation of PTx-loaded MPEG-PLGC films which affect to adjust the rate of release through to adjust the biodegradation rate. In this study, we prepared PTx-loaded methoxy-poly(ethylene glycol)-poly(ε-caprolactone-co-L-lactide-co-glycolide). MPEG-PLGC were synthesized by ring opening polymerization of ε-caprolactone, L-lactide and glycolide with various compositions. Then the solvent casting method allowed for the preparation of films and in vitro study for up to 40 days. The amount of PTx was analyzed using a high performance liquid chromatography (HPLC). Depending on the experimental conditions, the release rate was increased when CL ratio decreased from 60 to 20% and concentration of PTx decreased from 5 to 1 wt%. Also Fluorescein contained in the film was confirmed the movement to the outer wall from inner wall of blood vessels. In conclusion, we confirmed that PTx-loaded MPEG-PLGC films will be able to use as drug carrier or drug eluting stent. |
