제목 Engineered 3D lung carcinoma microenvironment for screening therapeutics and exploring drug resistance mechanism
초록 Polymeric hydrogels are used in 3D tissue models due to their structural similarity to the native cellular microenvironments and tunable properties. Hydrogel-based 3D tumor models are new platforms that can mimic a variety of in vivo tumor environments. Herein, we report an engineered 3D tumor microenvironment to examine the mechanisms of drug resistance in non-small cell lung cancer (NSCLC) cells. We created engineered 3D lung carcinoma constructs by encapsulating drug-resistant human NSCLCs into the gelatin and poly (ethylene glycol)-based interpenetrating polymer network (IPN) hydrogels. The hydrogels were formed via horseradish peroxidase (HRP)-mediated oxidative crosslinking reaction. Utilizing this hydrogel system, we fabricated artificial tumor models with similar mechanical properties to native lung tissue and explored the drug response and resistance mechanisms of NSCLCs. In conclusion, our hydrogels can be used for screening anticancer drugs and exploring drug resistance.
