제목 Characterization of Gold Binding Peptide Discovered via Microfluidic Biopanning System
초록 Biopanning is the process of screening peptides having high affinity and specificity to a target material. Limitations of traditional biopanning methods, which include requiring large amounts of the target material and inconsistent multiple pipetting steps to remove the low-affinity peptides from the surface, have led to the development of microfluidic biopanning systems.  While various microfluidic biopanning systems have been reported, few have targeted inorganic materials. Here, we report on a microfluidic biopanning system using gold as the target material. Simple photolithography techniques are used to fabricate the device, having a size of 2.5 cm by 2.5 cm. The affinity of the gold binding peptide (GBP) is analyzed using surface plasmon resonance. In addition, molecular dynamics simulations are performed to elucidate the binding mechanism of the GBPs to the gold surface. The simulations also offer explanation for the distinct association and dissociation rates of the GBPs.
