Coronavirus-detectingapproaches have been developed involving quantitative PCR (qPCR) and directtargeting of specific protein. However, there have been a few reports on thedetection of the RNA-dependent RNA polymerase (RdRP). Here, we introduce anovel diagnosis methodology of COVID-19 by detecting the RNA-directed and denovo RNA replicable function of RdRP. By leveraging amplification of RNAtemplates, our approach provides a general route towardsthe detection of RNA virus infection via tailor-made RNA platform for RNAdependent RNA polymerization. Wedevised an RNA platform for RdRP-induced transcription (RPRIT) that includesRNA template which can be directly transcribed by RdRP. By utilizing RPRIT, thepresence of RdRP could be confirmed under the isothermal incubation withoutpolymerase chain reaction (PCR). This novel RdRP-detecting method can open anew route for rapid diagnosis of the RNA virus-infected patients. |
