In situ RGD conjugated-Tetronic based hydrogel as an injectable bio-mimetic scaffoldfor tissue engineering applications |

Recently, the development of ideal biomaterials for tissue engineering has been focused on the design of bio-mimetic materials that can mimic the natural environments in the extracellular matrix. Arg-Gly-Asp (RGD) sequence, the signaling domain derived from fibronectin and laminin, has been commonly used for preparing the bio-mimetic scaffolds. In this study, in situ RGD-conjugated Tetronic-tyramine (Tet-TA) hydrogels was prepared via enzymatic reaction. Tet-TA hydrogel was rapidly formed and GRGDGGGGGY peptide was conjugated simultaneously in the presence of horseradish peroxidase and H2O2. In vitro cell study demonstrated that in situ conjugated RGD peptides with glycine and tyrosine residues facilitate the enhanced attachment of a variety of cells. Thus, in situ conjugation of the RGD peptide to injectable hydrogel via enzymatic reaction may be an efficient tool to prepare injectable bio-mimetic hydrogels for tissue engineering applications. |
