제목 | Effect of lactide/glcolide monomers on release behaviors of gentamicin sulfate-loaded PLGA devices for antibiotic local delivery system |
초록 | Gentamicin sulfate (GS), a highly water soluble antibiotic of the aminoglycoside group, is widely used for the treatment of serious infections such as osteomyelitis and the prevention of secondary infections after orthopedic surgery. GS/PLGA wafers and powders were prepared by direct compression method and melt extrusion method, respectively. For the desirable drug release patterns and removal of lag periods which frequently might show in the PLGA devices, lactide/glycolide monomers were introduced since they have non-toxic and biodegradable property in in vitro/in vivo. We have investigated the effects of sizes and additives. In vitro study was performed in pH 7.4 phosphate buffered saline (PBS) at 37℃. Drug contents and release amounts of GS were analyzed by high performed liquid chromatography (HPLC). Morphological evaluation of the devices was accomplished by scanning electron microscope (SEM). Moreover, differential scanning calorimetry (DSC) and gel permeation chromatography (GPC) were used for their characterization. From the result of in vitro release test, we could obtain the desirable drug release patterns without lag periods from the prepared GS/PLGA wafers and powders containing lactide/glycolide monomers.
