Size-Exclusion Chromatography Coupled with Small-Angle X-Ray Scattering on the 4C SAXS Beamline at Pohang Light Source II |

Small-angle X-ray scattering (SAXS) is now a popular technique in the field of structural biology for characterization of biological macromolecules in solution. However, the SAXS technique is associated with some difficulties and challenges, which can be alleviated by the combined use of size-exclusion chromatography (SEC) and SAXS (SEC-SAXS). Here, we describe the most recently installed SEC-SAXS equipment at 4C SAXS beamline at Pohang Light Source II (PLS II) in Korea. We also carried out SEC-SAXS measurements on model proteins to evaluate the performance of this system. Our study demonstrates that the current SEC-SAXS setup is suitable for the acquisition of reliable 3D structural models and for the preparation of high quality data. |
