제목 Fabrication of DNA/Protein G hetero-self-assembly Layer and its application to immunosensor
초록 The proteomics-based analytical tool consists of threebasic technologies. One of the most important points in the design of a protein chip is the proper choice of immobilization methode that will retain the activity of protein ligand molecule.
In this study, we investigated three cases. First case is self-assembled 11-mercaptoundecanoic acid (MUA) and protein G on gold surface by chemical binding. Second is self-assembled DNA A1 and on gold surface by chemical binding. Third is protein G and complementary DNA layer on mixed self-assembled MUA and DNA A1 on gold surface. This is for the steric effect. And we compared the efficiency of 3 cases as the immunosensor through the antigen-antibody interaction. Bovine serum albumin (BSA) and anti-BSA are used as antigen and antibody. The effect of 3 cases were investigated surface plasmon resonance (SPR) and atomic force microscopy (AFM) and investigated the possibility to the immunosensor.
