Quantitative analytical protocol for endogenous estrone in human breast cancer using MALDI-MS |
초록 |
Estrone is a major steroid hormone in women and is known to have a high correlation with the incidence of postmenopausal breast cancer. In this study, we develop a highly sensitive quantitative platform based on mass-assisted laser desorption/ionization mass spectrometry (MALDI-MS) to analyze the absolute amount of endogenous estrones in various clinical specimens. Derivatization of endogenous estrone has resulted in excellent linearity (R2> 0.99) and improved sensitivity platform (quantification limit: 11 fmol). In addition, the absolute amount of endogenous estrone could be confirmed by using deuterated estrone as an internal standard substance in the cells of the breast cancer cell line MCF-7 (34 fmol / 106 cells). Finally, we have successfully monitored the changes in the level of metabolic expression of estrogen (17.7 fmol / 106 letrozole treated cells) in MCF-7 cells treated with aromatase inhibitors via a MALDI-MS based quantitative platform. |
