Optogenetic Stimulation of Cortical Areas through Flexible Light-Emitting Diodes for Behavioral Control of Mouse |

The microstimulation of certain neural areas on the brain is one of the credible ways for induction of functional and complex behaviors of animals. Recently, advanced biomedical tools utilize flexible optoelectronic devices combined with optogenetic mouse models to facilitate spatiotemporal investigation of specific brain activities. However, most conventional applications are limited to activation of small functional areas using blue-light driven channelrhodopsin. In this work, we introduce flexible vertical light-emitting diodes (VLEDs) using AlGaInP to perturbate specific functional regions of mouse cortex. Micro-scaled LEDs effectively compress the conductive balls dispersed in anisotropic conductive film (ACF) resulting in emission of red light with high optical power density, which enables stimulating motor neurons deep below layer III under the brain surface. Selective illumination of pulsed red light from f-VLEDs induces synchronized electromyogram (EMG) signals and mouse body movements. The expression of chrimson, red-shifted channelrhodopsin, facilitates red-light excitation of targeted functional regions of motor cortex. This work opens new opportunities for cortical mapping, resulting in exploration of the interactions between motor areas in the mouse brain. |
