A Study of Electrocatalytic Reductive Amination for the Effective Enantioselective Synthesis |

The construction and functionalization of carbon-nitrogen bonds have been important tasks of chemical synthesis. Particularly, amines are basic structural units that constitute various natural products, pharmaceuticals, and agrochemicals. To prepare an amine, the most effective method is reductive amination as it exhibits high chemo-selectivity. In this study, we developed a new strategy to catalyze the reductive amination process. Starting with benzylic ketone and aliphatic amine, imine was prepared, which was further converted into the corresponding amine via electrochemical reduction by using Pd on carbon as electrocatalyst. As a result, about 20-40% Faradaic efficiency was obtained. We then tried to reveal the reaction mechanism from electrokinetic studies. We hypothesized that amino acid-based imines would be absorbed on Pd surfaces at certain configurations, inducing enantioselective synthesis of amines. We believe that our study will demonstrate an electrochemical pathway of reductive amination and suggest a possibility of a chiral additives-free electrochemical asymmetry synthesis. |
