Material characterization and In-Vitro biocompatibility and In-Vivo study of HAP scaffolds with In-vivo deposited Extra Cellular Matrix (ECM) for Bone Tissue engineering |
초록 |
This study aims to produce a sponge scaffold with inherent extracellular matrix (ECM) for bone tissue regeneration without employing the usual hydrogel coating or loading method. To incorporate the ECM to the HAp scaffold, samples of the scaffolds were implanted in the subcutaneous pockets of rats and fibrous tissue was allowed to propagate for up to three weeks. Implanted samples were then extracted and decellularized using a combination of 1% sodium dodecyl disulfate (SDS) and sonication. Material properties such as compressive strength and porosity were observed from samples after extraction and after decellularization. Morphology of the extracted and decellularized samples was also observed through scanning electron microscopy and coonfocal microscopy while histological sections of extracted and decellularized HAp scaffolds were compared with the original unmodified scaffold through histological staining. In vitro biocompatibility of the decellularized constructs was determined by seeding MC3T3-E1 pre- osteoblast cells and cell proliferation was observed using MTT assay and cell adhesion behaviour through confocal microscopy one, two and three weeks after seeding. Based on material characterization and in-vitro study results, 2 week decellularized HAp scaffols was chosen to implant in 11 mm radius defect of rabbit with 1 and 4 weeks observation period. New bone formation was evaluated through micro-CT scanning and histological analysis |
