We developed a coacervate (Coa)–mediated tumor cell lysate (TCL) delivery platform to strengthen the immune system against colon cancer. TCL from colon cancer, with multi-epitope efficacy, could induce effective activation of dendritic cells (DCs), as a specific tumor-associated antigen(TAA). However, TCLs are unstable and easily degraded after entering the body. Therefore, TCL was encapsulated in Coa for effective exogenous delivery with maintaining TCL stability and escaping in vivo degradation. Coa is self-assembly microdroplet composed of a biodegradable mPEGylated poly(ethylene arginylaspartate diglyceride) cation and counterpart heparin anion in an aqueous environment. About 90% of TCL encapsulation efficiency and sustained release pattern were obtained. Coa-mediated TCL delivery exhibited specific antigen presentation of DCs, and subsequent T cell activation. Therefore, our Coa system could be utilized TCL delivery platform for effective immune cell activation. |
