Nanoparticle Based on L-Tyrosine Polyurethanes for DNA Delivery |

A biodegradable polyurethane has been developed using L-tyrosine (LTU2a). LTU2a was designed to degrade through hydrolysis and enzymatic mechanisms since the backbone of the polymer contains a peptide bond. Using water-in-oil-in-water emulsion, nanoparticles encapsulating complexes of DNA and linear polyethylenimine LPEI or bovine serum albumin labeled with fluorescein have been fabricated. In addition, their surfaces have been converted with PEG. The morphology is spherical. After 14 days, 15 ng of DNA per mg of nanoparticles have been released from the LTU2a nanoparticles. Furthermore, a successful uptake of LTU2a nanoparticles by the hepatic cells has been confirmed with confocal laser microscope. These characteristics are ideal for drug-delivery systems designed to transport and release drugs in the cytoplasm. Therefore, LTU2a nanoparticles could be a promising for gene-delivery applications that require sustained release of DNA for the duration of a cell’s lifetime. |
