Although the metal binding ability of tryptophan is well known, it is relatively less known that it has unique quenching ability to fluorophore. We synthesized a fluorescent probe based on the natural amino acid with 4-dimethylamine-7-sulfonyl-2,1,3-benzoxadiazole fluorophore. This fluorescent chemosensor designed for detecting Hg(II) in aqueous solutions using dual role of tryptophan at higher wavelength. The probe includes tryptophan that acts as a ligand for heavy metal ions, especially mercury ion in this case, and quencher for fluorophore by photoinduced electron transfer (PET) process. In aqueous buffered solution, the probe showed highly selective turn-on response to mercury ion among 16 other metal ions. After the probe bound with mercury ion, it exhibited sensitive turn-on fluorescent response and color change with red shift. The cell penetration test conducted with ABD-W and it can measure concentration of mercury ion in the live cells effectively. |
