제목 | Evaluation of different alcohol dehydrogenases for bio-butanol production |
초록 | Bio-butanol is an attractive chemical that can be used as an alternative oil-based transport fuel and precursor of various chemical compounds. In general, bio-butanol could be produced by the reversed β-oxidation pathway constructing 4-5 genes. Among genes related to the pathway, alcohol dehydrogenase (adh) known for terminal enzymes is a significant enzyme. In order to evaluate adh in-vivo, butanol producing E. coli platform containing the acyl-CoA transferase gene (act, converting carboxylic acid into acyl-CoA) derived from Megasphaera hexanoica MHT was constructed. After that, four adhs (adhE, adhE2 and PduP-AdhA) were inserted and expressed in the strain, respectively. Productivities of four adhs were compared for performance evaluation. As a result, ranks for productivity showed like follows, PduP-AdhA>adhE>adhE2. (1.7 g/L/h, 1.1 g/L/h, 0.8 g/L/h, respectively) Finally, it is expected that these studies can affect increase of butanol production based on biological process. |
