Preparation and anticancer effect of pegylated-chlorambucil prodrug nanoparticle on the based on drug delivery system |
초록 |
Chlorambucil (CLB), used as therapeutic agent for leukemia, was recently demonstrated to anticancer effect against various cancer cells. However, its utilization has been limited due to MDR effect, high toxicity and low solubility against water. To overcome these obstacles, we synthesized pegylated-CLB by using coupling agents. Their chemical structure was analyzed by 1H-NMR and UV-vis spectrophotometer. To demonstrate a dispersity of them against water, transmittance was measured by using UV-vis spectrophotometer. Their particle size and morphology were analyzed by DLS and TEM. Moreover, cytotoxicity and anticancer effect were evaluated by using MTT assay. In addition, their cellular uptake against cancer cell lines was observed by using fluorescence microscopy and flow cytometry. These results suggest that they can use as anticancer agent.
