PET molecular Imaging study via Bioorthogonal F-18 labeling method |

F-18 labeling reaction of bioactive molecule via click reaction has been widely used to prepare F-18 labeled molecular imaging probes. In particular, bioorthogonal strain-promoted alkyne-azide cycloaddition (SPAAC) reaction has received much attention as an alternative conjugation method for radiolabeling bioactive molecules. In this presentation, I introduce the efficient mesoporous silica nanoparticles (MSNs) based pretargeting PET imaging protocol via the SPAAC reaction using F-18 labeled azide radiotracer in a live-body of tumor-bearing mice. I also introduce an efficient cell-tracking tool using PET system. From consideration of the fact of homing of macrophage to tumor, the tracking of RAW 264.7 cells using PET system in tumor-bearing mice was designed as a model of cell tracking via in vivo F-18 labeling reaction based on the bioorthogonal SPAAC ligation in the macrophage cells loaded by ADIBO-MSNs on tumor region together with 18F-azide in living subject. |
