제목 | Identification of cytosolic proteins of Streptomyces peucetius ATCC27952 using2D-gel electrophoresis and mass spectrometry |
초록 | Proteomics relying on two-dimensional polyacrylamide gel electrophoresis followed by protein identification with mass spectrometry is a promising experimental tool for physiological studies of microorganism. Streptomyces peucetius ATCC27952 is a doxorubicin(used broadly in chemotherapy) producing microorganism, and industrially useful. Here we report our own 2D reference map of Streptomyces peucetius ATCC27952 constructed by using isoelectric focusing followed by two-dimensional polyacrylamide gel electrophoresis. Protein identification was carried out by peptide tandem mass spectra using u-LC ESI-Tandem mass spectrometry. The two-dimensional gel pattern of proteins generated from any given cell system provides a fingerprint that is unique to those cells. This 2D reference map is applied to the study of antibiotics production regulation and doxorubicin resistance mechanism. |
