Development of high-throughput screening method for rapid selection of highly photo-inducible Haematococcus mutant |

For enhancing astaxanthin production from Haematococcus pluvialis, various efforts have been made to find astaxanthin-hyperproducing mutants. However, the development and actual exploitation of high-throughput screening for selecting the improved strains has not been common in H. pluvialis research field. Herein, we propose a novel approach for simple and rapid selection of highly photo-inducible Haematococcus mutants’ library through serially combined method of azide-based colorimetric assessment and oil-based astaxanthin quantification. Azide promptly triggered astaxanthin biosynthesis in H. pluvialis within 3 days by imposing artificial oxidative stress and oil-based extraction provided reliable and fast analytic method of the astaxanthin content without complex pretreatment. This universal screening method which accelerates both upstream and downstream of astaxanthin quantification would be useful for isolating the highly astaxanthin-producing microalgae. |
