In this study, we developed injectable, thermosensitive, biodegradable poly(organophosphazene) hydrogel system without extra implant or surgery. The synergy effect on bone formation by both BMP-2 and bisphosphonate was invesigated because bisphosphonate have the ability to enhance BMP-2’s osteogenic activity by the inhibition of osteoclastic bone resorption. The characterization of hydrogel was measured in terms of NMR, viscosity, in vitro cytotoxicity, and in vivo degradation test. BMP-2/hydrogel was assembled since a strong ionic interaction between negative charged carboxyl group in poly(organophosphazene). in vitro BMP-2 release test showed a sustained release pattern during 42 days without an initial burst release. We compared the bone formation test with the existence bisphosphonate or not in BMP-2 mixed hydrogel on ectopic site. Results demonstrated that the mice injected with BMP-2 and bisphosphonate showed significantly increased bone volume compared to the BMP-2 only group. |
