Globally, at least 2.8 million adults die each year as a result of being overweight or obese. Therefore, the development of efficient lipid inhibitor for the biological effects of obesity is highly needed. In this study, we described a simple and facile method to generate uniform microwells poly(dimethyl siloxane) (PDMS) microstamps through micro-molding for efficient and reliable cell patterning of adipocyte differentiation. Here, we demonstrated Orlistat as a anti-obesity compound showed the lipid droplet inhibition activity on 3T3-L1 cell patterning platform. This platform can be used for the real-time monitoring of cell proliferation and differentiation in live cell-based assays. The array technology described here may readily be expanded to discover a wider variety of anti-obesity agents on one-pot cell array chip. |
