Development of Human Cardiac Organoids for Drug Screening |

Drug-induced cardiotoxicity has been one of the major concerns that lead to drug withdrawals from markets and early discontinuation of promising drug candidates. Currently, in vitro hERG ion channel assay and in vivo telemetry for QT screening in conscious animals are used as standardized assays for cardiotoxicity. However, there are several limitations for the absence of physiological environment in the hERG assay and the ethical issues and interspecies differences in animal studies. We have developed cardiac organoids using human induced pluripotent stem cells. In addition, we also developed a comprehensive toxicity testing platform based on electrophysiological and image-based analysis for an in vitro alternative to animal testing. The human cardiac organoid-based toxicity testing platform could be more reliable to evaluate proarrhythmic risk than previous non-clinical assays. |
