Stem cells usually are detached from the cell culture platform for harvest and transplantation. In the conventional detachment process, stem cell-based therapy against ischemia disease is negatively affected by proteases. Herein, we developed an alternative stem cell detachment method with pH-sensitive biodegradable polymer; cell detachable polymer (CDP). Without any adverse effect on growth, almost all of human adipose tissue-derived stem cells (hADSCs) on CDP-coated surface were detached under pH 6.0 condition within a short period. Compared to conventional enzymatic cell detachment, hADSCs detached from CDP-coated surface showed similar cellular properties. The in vivo results of mouse hindlimb ischemia model indicated that transplant of detached hADSCs with both enzymatic and CDP-coating methods showed analogous therapeutic angiogenesis. In conclusion, CDP-coating method were applied to a pH-responsive cell detachment system as a non-enzymatic method for stem cell-based therapy.. |
