Herein, pH/redox responsive cationic polymer dot (CD) was synthesized for controllable photothermal effect using off/on fluorescent system to control therapeutic agent’s release. The disulfide crosslinked polyethyleneimine (bPEI) was carbonized and conjugated with folic acid (FA) and partially ionic complexed with indocyanine green (ICG), resulting in bPEI-based CD (ICG-CD). ICG-CD was internalized into cancer cells via FA-receptor and endocytosis in OFF-state, and fluorescence partially recovered in high GSH, then internalized into acidic endosomes to proceed complete fluorescence restoration. This tumor-selective degradability also allowed controllable photothermal therapy under NIR irradiation, improving anticancer efficiency. As a result, ICG-CD displayed potential in tumor targeting, intracellular imaging, and controllable therapeutic delivery via fluorescence off/on assay. |
