Polydiacetylene(PDA) liposomes have unique properties that allow liposomes to change color and emit fluorescence in response to external stimulis. In here, we demonstrate an antibody-based biosensor using PDA liposomes for detection of allergy diagnosis by human immunoglobulin E(hIgE). In order to detect target hIgE, the surface of immoblized PDA liposomes were functionalized with hIgE monoclonal antibodies and the fluorescent signals were detected after antigen-antibody immunoresponse. Fluorescent signals were slightly increased depending on the target protein concentration when polyclonal antibody conjugated with horseradish peroxdiase(HRP) were used after primary immunoresponse. After secondary immunoresponse,HRP-biocatalyzed oxdiation of 3,3’-diaminobenzidine(DAB) in the presence of H2O2 formed precipitation of an insoluble product. As a result, a PDA liposome biosensor could detect effectively the hIgE and the sensitivity was evidently increased higher than the primary response. |
