Herein, we demonstrate an antibody-based biosensor using PDA liposomes for detection of allergy diagnosis by human immunoglobulin E (hIgE). In order to detect target hIgE, the surface of immoblized PDA liposomes were functionalized with hIgE monoclonal antibodies and the fluorescent signals were detected after antigen-antibody immunoresponse. However, fluorescent signals were slightly increased depending on the target concentration when polyclonal antibody conjugated with horseradish peroxdiase(HRP) were used as an amplifyling agent after primary immunoresponse. After secondary immunoresponse, HRP-biocatalyzed oxdiation of 3,3’-diaminobenzidine(DAB) in the presence of H2O2 formed precipitation of an insoluble product. The precipitation strongly stimuli on PDA liposomes due to their weight and pressure, thus fluorescent signals have increased dramatically. |
