제목 Overcoming the Multidrug Resistance with Ultrasound Activated Chemotherapy and Photodynamic Therapy
초록 Multiple drug resistance is a critical barrier in drug delivery system which decrease delivery efficiency. In this study, we apply photodynamic therapy with chemotherapy using albumin nanoparticles with doxorubicin and microbubbles containing Chlorin e6 (DOX-NPs/Ce6-MBs) to overcome the MDR and maximize the therapeutic effect. Microbubbles, which can increase delivery efficiency by cavitation effect, are used as carrier of Chlorin e6. Reactive oxygen species generated by PDT had functional destruction effects on efflux pumps. In the group treated with NPs-MBs without ultrasound and laser, cells showed cell viability of 90.5% compared to the control group, whereas 47.7% cell viability was observed in the group irradiated with ultrasound and laser. As a result, DOX-NPs/Ce6-MBs improved chemotherapy and PDT by ultrasound trigged local delivery and over-deliver of nanocarriers by sonoporation, and the efflux pumps were inhibited by the reactive oxygen species to prevent drug release.
