Graphene Oxide-Peptide Hybrids for Optical Detection of Matrix Metalloproteinase-2 by Fluorescence Turn-on Response |

Graphene oxide (GO) is a novel material that has been rapidly growing in the field of material science with its remarkable physicochemical properties. Especially, photoluminescence of GO suggest that it has great potential for optical sensing area with photo-stability and unique photoluminescence (PL) property appearing in the visible and near-infrared region. In addition, GO based sensing platform have attracted great attentions due to the featured properties of GO, such as biocompatibility, and chemical affinity to biomolecules. Herein, we present GO PL-based optical biosensor consisting of a luminescent GO donor covalently linked to a peptide-quencher complex for the simple, rapid and sensitive detection of specific proteases. The designed GO-peptide hybrid was able to detect specific proteases, chymotrypsin and matrix metalloproteinase-2 (MMP-2), by the “turn-on” response of quenched GO fluorescence after proteolytic cleavage and releasing the quencher. Furthermore, the GO-peptide-quencher hybrid successfully monitored MMP-2 secreted from living cells with high sensitivity. |
