Development of immunity-activating interfaces by surface modification of pH-sensitive polymers onto lipid membranes and their application to cancer immunotherapy |

For establishment of cancer immunotherapy, efficient antigen carriers that can deliver antigen into cytosol of dendritic cells (DCs) and activate DCs are required. In this study, intracellular antigen delivery systems were developed using liposomes modified with poly(carboxylic acid) derivatives which become hydrophibic at weakly acidic pH and destabilize lipid membrane. pH-Sensitive polymer-modified liposomes delivered antigenic proteins into cytosol of DCs and the administration of liposomes to tumor-bearing mice induced antigen-specific cellular immune responses, which achieved the regression of tumor. In addition, inclusion of molecular adjuvants such as cationic lipids, toll like receptor ligands or bioactive polysaccharides improved the activation property of DCs by pH-sensitive polymer-modified liposomes, immunity-inducing effects and therapeutic effects against tumor-bearing mice. Therefore, these liposomes are promising antigen delivery carriers for cancer immunotherapy. |
