Immobilization of protein in 3D mesoporous TiO2 matrice for cancer biomarker detection |
초록 |
Three-dimensional (3D) substrate can provide the increased surface area for improved modalities of protein adsorption and performance of the assay. In this work, we successfully fabricated TiO2 nanoporous substrates which provide an enhanced surface area twofold or more compared with planar substrate to enhance the limit of detection of target biomolecules with enhanced sensitivity. TiO2 nanoporous substrates can be prepared using poly(vinyl chloride) (PVC)-g-poly(oxyethlynene methacrylate) (POEM) graft copolymers as a structure-directing agent. For confirming the sensing capability of our system, we immobilized specific antibodies onto TiO2 substrate which can be used for detecting prostate-specific antigen (PSA) with antigen-antibody interaction. Our system can be exploited as PSA detectable immunoassays which have great potential in early prostate cancer diagnostics and therapeutic monitoring. |
