Prepration of cross-linked peptosome via reversible disulfide bonding |

One fundamental problem with block copolymer micelles is their spontaneous dissociation at concentrations below their critical micelle concentration (CMC). Shell or Core cross linking method is general method to overcome this problem. We synthesized the poly(2-hydroxyethyl asparamide)-graft-Oligo(cysteine)(PHEA-g-OCys) amphiphilic graft copolymers which is the synthetic peptide. Because the oligocysteine is hydrophobic, the PHEA-g-OCys formed veslicle like aggregates in aqueous solution. Thiol residues of oligocysteine were easily cross-linked by oxidation reaction and formed disulfide linkages. The molecular structure of PHEA-g-OCys was measured by 1H NMR spectroscopy. The fixed vesicle structure of cross-linked PHEA-g-OCys was measured by TEM and AFM. Average diameter and size distribution of micelles were measured by DLS. Oxidation reaction of thiol residues was monitored by the ellman’s method. |
