Cancer Targeted Delivery of Functionalized Adenovirus via the Folate Receptor |

Surface functionalized adenovirus (ADV) carriers with low-molecular chitosan (LMC), polyethyleneglycol (PEG), and folate (FA) were fabricated by electrospinning for intravascular injection (IV). ADV was encapsulated with LMC by using electrospinning. To confirm LMC coating of ADV, surface charges were measured by ζ-potential. Morphology was observed by TEM. LMC was prepared by cross-linking using sodium tripolyphosphate (TPP). In case of linking concentration affected the cell viability. In vitro studies in cells line U343, have revealed that coated ADV association effect. This result was corroborated by FACS. PEG-FA synthesized in advance was conjugated with ADV-cLMC. The surface functionalized ADV carriers also had cancer targeting moiety due to FA modification. The synthesis was confirmed with 1H NMR and FTIR. Cell viability was according to FA concentration measured by FACS. ADV-LCPF was cytotoxic test for a folate receptor over expressing cell line, KB cell. |
