The interdiffusion of nano-objects within layer-by-layer (LbL) multilayered films has recently received great attention due to its unique molecular transport phenomena. In particular, the use of weak polyelectrolyte enables us to construct films that are responsive to the variation of external pH conditions. In the present study, we have established a novel platform based on the protein assembly within polymeric thin films through the interdiffusion of precursors of amyloid fibrils. κ-Casein and poly(acrylic acid) (PAA) were used as an amyloid precursor and a weak polyelectrolyte interacting with the proteins in this model system. With the isoelectric point of κ-casein around pH 4.5, the LbL films were constructed at pH 3.0 and the films were then subject to incubation in pH 8.0 at 80 ℃, and this pH value altered the molecular interactions between κ-casein and PAA such that amyloid fibrils were formed within the multilayer films. |
