제목 A Study on the Substrate Binding of Poly(hydroxybutyrate) Depolymerase by 2D IR COS
초록 Poly(hydroxybutyrate)(PHB) depolymerases have the substrate binding domain (SBD) which binds onto the surface of poly[(R)-3-hydroxybutyrate](P(3HB)) film. In this study, we investigated the binding between SBD of PHB depolymerases and PHB surface using two-dimensional infrared correlation spectroscopy (2D IR COS). Variation in water uptake in SBD on the PHB film was employed as a perturbation to generate a gradient in IR spectra of SBD and PHB. 2D IR COS simplified the overlapped peaks attributed by side chain residues of SBD and enhanced the spectral resolution. The cross peaks in the 2D correlation synchronous spectrum indicated the amino acid residues with hydrophilic side chains, such as asparagines and glutamines, have interactions with carbonyl groups in PHB, and positively charged residues also seem to be involved in the interactions. 2D IR correlation spectra of SBD immobilized on PHB were compared to the spectra of the SBD only solid film.
