Sequential protein delivery using a microsphere/hydrogel combination system for therapeutic angiogenesis |

Cardiovascular disease causes high mortality rate to date, and delivery of therapeutic protein drugs in a minimally invasive manner may reduce the rate. We hypothesized that protein drugs such as heat shock protein (HSP) and vascular endothelial growth factor (VEGF) could be sequentially released from a microsphere/hydrogel combination system, and this system could be useful for therapeutic angiogenesis. HSP protects cells from cellular apoptosis under the hypoxia condition, and VEGF is a potent angiogenic molecule that has been frequently used for therapeutic angiogenesis. The sequential release of TAT-HSP27 (HSP fused with transcriptional activator) and VEGF from the system was achieved in vitro and significantly enhanced new blood vessel formation in ischemic hindlimb mouse model. This approach to controlling the release of various protein drugs in a sequential manner may be useful in treatment of ischemic disease, as well as in many drug delivery applications. |
