Reduction-sensitive monoolein cubic phase nanoparticle (cubosome) containing epidermal growth factor (EGF) was prepared by inclosing alginate and cystamine in the cubosome water channel. The alginate and cystamine contained in the cubosome water channel is crosslinked by electrostatic attraction. The morphology of cubosome was observed by atomic force microscopy. The release of EGF included in the cubosome was investigated according to the concentration of dithiothreitol (DTT, reducing agent). In the presence of DTT, the release of EGF was markedly increased at higher DTT concentration because the disulfide bond of cystamine was broken. The skin permeation of free EGF and EGF containing cubosome suspension was compared through the artificial skin attached to the diffusion cell.
