제목 One-pot Biocatalytic Synthesis of Nylon Monomers from Cyclohexanol Using Escherichia coli-based Cascade
초록 The structural moieties 6-aminohexanoic acid (6-AmHA; 6) and ε-caprolactam are widely used for synthesizing polyamides. Nylon-6 and nylon-6,6 represent approximately 90% of the total nylon produced daily worldwide, amounting to nearly 7.0 million tons annually. We envisaged an in vivo multienzyme cascade designed for the production of nylon building blocks using an Escherichia coli-based consortia system with the following objectives: 1) a highly atom-efficient and self-sufficient redox network using cheap substrate and co-substrates, 2) self-sufficient modularization of the reaction pathway without any auxiliary enzymes for cofactor recycling, and 3) distribution of the cascade into different modules (cells) to manage the protein expression burden. The complete pathway was distributed into three reaction modules. Herein, a cell-based modularization strategy was applied to produce 6-aminohexanoic acid and ε-caprolactam from cyclohexanol.
