Fabrication, AFM Imaging and Electrical Signal Analysis of Si-Nanowire Field Effect Transistor Aptamer Biosensor |

We report the fabrication, AFM imaging and electrical signal analysis of Si nanowire field-effect transistor (SiNW-FET) aptamer biosensor. Surface modifications were carried out using 3-aminopropyl diethoxysilane and succinic anhydride to introduce amine and carboxyl groups on Si-NW. Aptamers with 5’-end amine groups were chemically grafted on the surface-modified Si substrates through the amide bond formation. Atomic force microscopic analysis confirmed the successful immobilization of aptamers on Si nanowire and the following protein binding to the aptamers. Furthermore, fluorescence micrograph clearly visualized the attached protein to aptamers immobilized on Si-NW. The Si-NW aptamer biosensor was successfully applied for the real-time detection of electronic signals during and after protein binding. |
